| Beschreibungen: | Trotz zahlreicher therapeutischer Vorstöße wird akute myeloische Leukämie (AML) mit hohen Rückfallraten und schlechten Überlebensraten assoziiert. Die 5-JahresÜberlebensrate für Erwachsene / Kinder beträgt 65 % / 30 %. In präklinischen Studien wurden Histondeacetylasen (HDAC) als effektive epigenetische Ziele in AML identifiziert. Allerdings zeigten unselektive HDAC-Inhibitoren (HDACi) nur geringe antileukämische Effekte und schwerwiegende Toxizität. Daher ist ein besseres Verständnis des Mechanismus von HDAC-Inhibitoren und die Identifizierung von synergistischen Therapeutika notwendig. Innerhalb der HDAC-Protein-Familie hat HDAC6 wegen seiner Struktur, welche eine selektive Inhibition erlaubt, ein besonderes Potenzial. HDAC6 reguliert eine Vielzahl krebsrelevanter Stoffwechselprozesse wie die Hitzeschockreaktion, Immunflucht, Zellmobilität und Autophagie. Da die Auswirkungen von HDAC6-Inhibition jedoch äußerst kontextabhängig sind, müssen sie systematisch innerhalb der verschiedenen Leukämie-Subtypen charakterisiert werden.
Diese kumulative Dissertation umfasst vier wissenschaftliche Publikationen. Drei der Studien fokussieren sich auf die Charakterisierung von klassenspezifischen HDACi in einer breiten Auswahl von Leukämiezelllinien. Leitstrukturen mit verbesserten antileukämischen Effekten wurden hinsichtlich ihrer Isoenzymselektivität und Fähigkeit zur Apoptose- oder HDAC-Substrat-Acetylierungsinduktion charakterisiert. Aus diesen Bestrebungen gingen vier äußerst potente Klasse-I/IIb-HDACi (10h, 1g, 4d und 4m), ein HDAC6-präferierender Inhibitor (2b) und ein hochselektiver HDAC6-Inhibitor (10p) hervor. Jeder dieser Inhibitoren zeigte ein unterschiedliches Maß an synergistischer Interaktion mit dem Standard-AMLChemotherapeutikum Decitabin, allerdings waren 10h, 2b und 4d besonders effektiv. Die letzte Publikation fokussierte sich auf HDAC6-Inhibition in myeloischer Leukämie. Über CRISPR-Cas9 wurde der Verlust von HDAC6 modelliert, und der präferentielle HDAC6- Inhibitor Ricolinostat wurde für eine pharmakologische Kreuzvalidierung genutzt. Während die Zellen in vitro ungehindert wuchsen, war in vivo die Leukämiebürde reduziert. Eine Multi-Omik-Analyse via Proteomik, Secretomik, RNA-seq und ATAC-seq zeigte signifikante Veränderungen diverser Stoffwechselwege. Besonders lysosomale Proteine, wie der Tumorsuppressor RNase T2, waren angereichert in den untersuchten Zelllinien. Zudem sensibilisierte Ricolinostat AML-Zellen für CD8+-T-Zell-vermittelte Toxizität. Durch ein Hochdurchsatz-Medikamentenscreening konnte HDAC6 als ein Mediator für Resistenz gegen das Standard-AML-Chemotherapeutikum Cytarabin identifiziert werden. Basierend auf diesen Ergebnissen erscheint HDAC6-Inhibition als potenzielle Behandlungsmöglichkeit für myeloischer Malignome, jedoch bedarf es weiterer Forschung.Acute myeloid leukemia (AML) remains associated with high relapse rates and poor overall survival despite recent therapeutic advances. The 5-year survival rate is approximately 30% in adults and 65% in children. Preclinical studies have identified histone deacetylases (HDACs) as effective epigenetic targets in AML. However, clinical application of pan-HDAC inhibitors has been limited by modest antileukemic efficacy and substantial toxicity. This underscores the need for a more fundamental understanding of HDAC inhibitor (HDACi) mechanisms and the identification of synergistic therapeutic partners. Among the HDAC family, HDAC6 represents a particularly promising target due to its unique structural properties that permit selective inhibition. HDAC6 regulates multiple cancer-relevant processes, including the heat shock response, immune evasion, cell motility and autophagy. Given the context-dependent nature of these effects, systematic evaluation of HDAC6 inhibition across distinct AML subtypes is warranted. This cumulative dissertation summarizes the findings of four scientific publication. Three of these studies focus on the characterization of novel class-specific histone deacetylase inhibitors (HDACi) in a broad panel of leukemic cell lines. Lead compounds demonstrating superior antileukemic activity were further analyzed with respect to their HDAC isoenzyme selectivity and their capacity to induce apoptosis or hyperacetylation of HDAC substrates. These investigations identified four highly potent class I/IIb HDACi (10h, 1g, 4d, and 4m), one HDAC6-preferential inhibitor (2b), and one highly HDAC6-selective inhibitor (10p). All inhibitors exhibited varying degrees of synergistic activity with the AML standard chemotherapeutic agent decitabine, with compounds 10h, 2b and 4d showing the most pronounced effects. The last paper focuses specifically on HDAC6 inhibition in myeloid leukemia. HDAC6 loss models were generated using CRISPR-Cas9, while the HDAC6 preferential inhibitor ricolinostat was used for a pharmacological cross validation. While proliferation in vitro was unaffected, in vivo experiments showed a significant reduction in leukemia burden. A multiomics analysis through proteomics, secretomics, RNA-seq, and ATAC-seq identified major shifts in a multitude of pathways. Notably, lysosomal proteins, such as the tumor suppressor RNase T2, were highly enriched across a broader panel of cell lines. Furthermore, ricolinostat induced susceptibility of AML cells to CD8+ T cell-mediated cytotoxicity. Highthroughput drug screening revealed the specific role of HDAC6 in resistance to the common AML chemotherapeutic cytarabine. Overall, specific targeting HDAC6 may serve as a viable strategy in the treatment of myeloid malignancies, warranting further investigations. | |||||||
