| Beschreibungen: | Keimzelltumoren (KZT) sind die häufigste solide maligne Erkrankung des Mannes im Alter von 20 bis 44 Jahren. Sie können histologisch in Seminome (SE) und Nicht-SE unterteilt werden. Mit einer relativen 5 Jahres Überlebensrate von 97 % haben sie unter den malignen Tumorerkrankungen eine gute Prognose. Allerdings ist die relative Überlebenswahrscheinlichkeit im Vergleich zur Allgemeinbevölkerung deutlich reduziert. Derzeit gibt es kaum Behandlungsalternativen für Cisplatin-resistente KZT, so dass 10 – 15 % der Patienten mit einem fortgeschrittenen KZT und 3 – 5 % aller Patienten mit einem KZT versterben. Tumoren stellen eine komplexe Struktur aus Tumorzellen, nicht-tumoralen Zellen (Fibroblasten, Immunzellen, Endothelzellen, u.a.) und Makromolekülen dar, dem sogenannten Tumormikromileu (TMM). Inzwischen ist es allgemein anerkannt, dass das TMM das Fortschreiten, die Metastasierung und das therapeutische Ansprechen von Tumoren beeinflusst und dies nicht nur von autonomen Prozessen der Tumorzellen abhängt. Allerdings ist das TMM in KZT noch unzureichend beschrieben. Deswegen war das Ziel dieser in vitro Arbeit eine umfängliche Charakterisierung des dreidimensionalen Wachstums von KZT-Zellen in Abhängigkeit von der Anwesenheit von TMM-Zellen durchzuführen. Hierfür wurden die häufigsten KZT-Subtypen (SE, Embryonalkarzinom (EK) und Chorionkarzinom (CK)) mit TMM-Zellen (Fibroblasten, Lymphozyten, Endothelzellen und Makrophagen) in der Hanging Drop Zellkulturtechnik ko-kultiviert und mikroskopisch untersucht. Um die KZT-Zellen mikroskopisch von den TMM-Zellen unterscheiden zu können, wurden die KZT-Zelllinien mit einem GFP-Plasmid und die TMM-Zelllinien mit einem mCherry-Plasmid transduziert.
Die EK-Zelllinien (NCCIT, 2102EP) und SE-Zelllinie (TCam-2) wiesen ein stabiles Wachstum auf. Sie waren sowohl in der Mono- als auch den unterschiedlichen Ko-Kulturen in der Lage ein kugelförmiges Sphäroid zu bilden. Im Gegensatz dazu wuchsen die CK-Zelllinien (JAR, JEG-3) flach und planar. Die Ko-Kultivierung mit Lymphozyten (JURKAT) und Fibroblasten (MPAF) führte jedoch ebenfalls zur Ausbildung eines kugelförmigen Sphäroids der CK-Zellen. Gleichzeitig zeigte sich eine verminderte Ausprägung der Lymphozyten in diesen Ko-Kulturen. Die Ergebnisse dieser Arbeit liefern neue Hinweise auf die Interaktion von KZT-Zelllinien und TMM-Zelllinien in der 3D-Zellkultur. Insbesondere die CK-Zelllinien scheinen eine ausgeprägte immunsuppressive Wirkung auf Lymphozyten auszuüben, was mit der geringen Effektivität von Immuncheckpointinhibitoren bei dieser Tumorentität einhergehen könnte. Die Beobachtung, dass Fibroblasten (MPAF) und Endothelzellen (HUVEC) in Ko-Kulturen mit KZT-Zelllinien ein besonders stabiles Wachstum zeigten, legt nahe, dass diese Zelltypen eine zentrale Rolle im TMM spielen. Insbesondere bei therapieresistenten CK könnten sie potenzielle Zielstrukturen für neue Behandlungsstrategien darstellen.Germ cell tumors (GCT) are the most common solid malignant disease in men aged 20 to 44. They can be divided histologically into seminomas (SE) and non-SE. With a relative 5-year survival rate of 97 %, they have a good prognosis among malignant tumor diseases. However, the relative probability of survival is significantly reduced compared to the general population. Currently, there are few treatment alternatives for cisplatin-resistant GCT, meaning that 10–15 % of patients with advanced GCT and 3–5 % of all patients with GCT die. Tumors are complex structures consisting of tumor cells, non-tumor cells (fibroblasts, immune cells, endothelial cells, etc.) and macromolecules, known as the tumor microenvironment (TMM). It is now widely accepted that the TMM influences the progression, metastasis, and therapeutic response of tumors and that this does not depend solely on autonomous processes of the tumor cells. However, the TMM in GCTs has not yet been adequately described. Therefore, the aim of this in vitro study was to comprehensively characterize the three- dimensional growth of GCT cells depending on the presence of TMM cells. For this purpose, the most common GCT subtypes (SE, embryonal carcinoma (EC), and choriocarcinoma (CC)) were co-cultured with TMM cells (fibroblasts, lymphocytes, endothelial cells, and macrophages) using the hanging drop cell culture technique and examined microscopically. In order to distinguish the GCT cells from the TMM cells microscopically, the GCT cell lines were transduced with a GFP plasmid and the TMM cell lines with a mCherry plasmid. The EC cell lines (NCCIT, 2102EP) and SE cell line (TCam-2) showed stable growth. They were able to form spherical spheroids in both mono- and different co-cultures. In contrast, the choriocarcinoma cell lines (JAR, JEG-3) grew flat and planar. However, co-cultivation with lymphocytes (JURKAT) and fibroblasts (MPAF) also led to the formation of a spherical spheroid of CC cells. At the same time, there was a reduced expression of lymphocytes in these co-cultures. The results of this study provide new insights into the interaction of GCT cell lines and TMM cell lines in 3D cell culture. CC cell lines in particular appear to exert a pronounced immunosuppressive effect on lymphocytes, which could be associated with the low effectiveness of immune checkpoint inhibitors in this tumor entity. The observation that fibroblasts (MPAF) and endothelial cells (HUVEC) showed particularly stable growth in co- cultures with GCT cell lines suggests that these cell types play a central role in TMM. They could represent potential target structures for new treatment strategies, particularly in therapy- resistant CC. | |||||||
