Testikuläre Keimzelltumoren (KZT) sind bei Männern im Alter von 14 - 44 Jahren die häufigste Tumorerkrankung. Obwohl die Prognose im Allgemeinen sehr gut ist, kann die Erkrankung aufgrund der Entwicklung von Resistenzmechanismen gegenüber der Standard-Cisplatin-basierten Chemotherapie tödlich verlaufen. Hier bietet die CXCR4 / CXCR7 / CXCL12-Achse einen neuen Forschungsansatz für zusätzliche Therapiemöglichkeiten. Die Chemokin-Rezeptoren CXCR4 und CXCR7 sind G-Protein-gekoppelte Rezeptoren (GPCR, „G protein-coupled receptor“), welche durch ihren gemeinsamen Liganden, dem aus dem Tumor- Mikromilieu (z. B. von Fibroblasten) sezernierten Chemokin CXCL12, aktiviert werden. Folglich können nachgeschaltete Signalwege der MAP-Kinase, NF-κB, AKT / PKB oder auch JAK / STAT aktiviert werden. In vielen Tumoren (u.a. in KZT) sind Zellen CXCR4 und / oder CXCR7 positiv; z.B. beim Mamma-, Kolorektal- oder dem Prostatakarzinom ist bekannt, dass die CXCR4 / CXCR7 / CXCL12 -Achse eine Schlüsselrolle in der Proliferation, Migration, Angiogenese und Tumorinvasion spielt und mit einer schlechten Prognose einhergeht. Somit bietet diese Achse ein therapeutisches Ziel, welches als Medikament bereits für Non-Hodgkin-Lymphome mit dem CXCR4- Antagonisten „Plerixafor“ durch die europäische und amerikanische Arzneimittelbehörde zugelassen ist. Das Ziel der Promotionsarbeit war die Aufschlüsselung der molekularen Mechanismen der aus Fibroblasten sezernierten CXCL12-Stimulation in testikulären KZT; insbesondere sollte erforscht werden, welche Signalwege durch CXCL12-Stimulation aktiviert werden, da dies in testikulären KZT unzureichend bekannt ist. Zudem sollte herausgefunden werden, welchen Einfluss die Achse auf die Migration und Proliferation von Keimzelltumor-Zellen hat und ob CXCR4 und CXCR7 Ziele für neue Therapiemöglichkeiten darstellen. Insgesamt konnten durch Western-Blot-Analysen, PCRs und Human-Phospho- Kinase-Arrays in dieser Arbeit mehrere Signalwege in KZT, welche unter dem Einfluss der CXCR4 / CXCR7 / CXCL12-Achse stehen identifiziert werden (z.B. p53). Zusätzlich wurde mittels Zellviabilitäts-, Apoptose- und Zellzyklusanalysen ein CXCR4-spezifisches Nanobody-Wirkstoff-Konjugat als mögliche therapeutische Option in KZT identifiziert.Summary (english) Testicular germ cell tumors (GCT) are the most common type of cancer in men aged 14–44. Although the prognosis is generally very good, the disease can be fatal due to the development of resistance mechanisms to standard cisplatin- based chemotherapy. Here, the CXCR4 / CXCR7 / CXCL12 axis offers a new research approach for additional therapeutic options. The chemokine receptors CXCR4 and CXCR7 are G protein-coupled receptors (GPCR) that are activated by their common ligand, the chemokine CXCL12, which is secreted from the tumor microenvironment (for example fibroblasts). Consequently, downstream signaling pathways of MAP kinase, NF-κB, AKT / PKB or JAK / STAT can be activated. In many tumors (including GCT), cells are CXCR4 and/or CXCR7 positive, such as breast, colorectal, or prostate cancer, the CXCR4 / CXCR7 / CXCL12 axis is known to play a key role in proliferation, migration, angiogenesis, and tumor invasion and is associated with a poor prognosis. This axis therefore offers a therapeutic target, which has already been approved as a drug for non-Hodgkin's lymphoma by the European and American drug authorities with the CXCR4 antagonist “Plerixafor”. The aim of the doctoral thesis was to break down the molecular mechanisms of CXCL12 stimulation secreted from fibroblasts in testicular germ cell tumors; in particular, the aim was to investigate which signaling pathways are activated by CXCL12 stimulation, as this is insufficiently known in testicular germ cell tumors. In addition, the aim was to determine the influence of the axis on the migration and proliferation of germ cell tumor cells and whether CXCR4 and CXCR7 represent targets for new therapeutic options. Overall, Western blot analyses, PCRs and human phospho-kinase arrays in this study identified several signaling pathways in GCT that are influenced by the CXCR4/CXCR7/CXCL12 axis (for example p53). In addition, cell viability, apoptosis, and cell cycle analyses identified a CXCR4-specific nanobody drug conjugate as a potential therapeutic option in GCT. | |||||||
