Beschreibungen:Feline immunodeficiency virus (FIV) and human immunodeficiency virus (HIV) are Lentivirus members within the Retroviridae family, sharing structural, genetic, and transmission similarities. FIV serves as a valuable model for studying HIV-induced AIDS. The host restriction factor APOBEC3 (A3) inhibits both HIV and FIV by introducing hypermutations into viral genomes. To counteract A3, HIV and FIV encode viral infectivity factor (Vif), which recruits Cullin-RING E3 ubiquitin ligases to degrade A3 through a proteasome dependent mechanism. Understanding Vif-A3 and Vif-Cullin interactions holds promise for HIV therapeutic development.
This study characterizes feline lentiviral Vif-mediated A3 degradation and the restriction activity of feline A3Z3 against FIV reporter viruses. Puma lentivirus (PLV) type 1695 Vif, identified as incapable of degrading feline A3Z3 but effective against A3Z2 and A3Z2Z3, highlights residue 201 as a key determinant for A3Z3 interaction. Mutation at this position in PLV1695 Vif explains its inability to counteract A3Z3. The 201YRF203 motif in PLV type B (PLVB) Vif, conserved across FIV domestic cat-, PLV type A (PLVA)-, and HIV-1 Vif, is essential for A3 interaction and degradation. Additionally, FIV and PLVA Vif degrade A3Z3 through both their N- and C-terminal regions, whereas PLVB Vif depends on its C-terminal, with the corresponding N-terminal inactive in A3Z3 degradation.
Regarding Vif-Cullin interactions, this study reveals differential binding of feline Vif proteins to CUL2 and CUL5. A conserved interface mediating CUL5 binding was identified in HIV-1 and feline Vifs. Notably, PLVB Vif recruits both CUL2 and CUL5 E3 ligase to degrade A3Z3. A novel interaction between HIV and feline Vif with CUL1 was discovered, although its functional relevance requires further investigation.
In summary, this work provides a novel perspective on the interactions between lentiviral Vif and host A3, as well as host Cullin proteins. These findings offer new avenues for uncovering the complex dynamics of lentiviral protein-host protein interactions.

Felines Immundefizienzvirus (FIV) und das Humane Immundefizienzvirus (HIV) gehören zur Gattung Lentivirus in der Familie Retroviridae und weisen strukturelle, genetische und Ähnlichkeiten in der Übertragung auf. FIV dient als wertvolles Modell zur Untersuchung der durch HIV induzierten AIDS-Pathogenese. Der Wirtsrestriktionsfaktor APOBEC3 (A3) hemmt sowohl HIV als auch FIV durch das Einfügen von Hypermutationen in virale Genome. Um A3 entgegenzuwirken, kodieren HIV und FIV den viralen Infektiositätsfaktor (Vif), der Cullin-RING-E3-Ubiquitin-Ligasen rekrutiert, um A3 in einem Proteasom abhängigen Mechanismus abzubauen. Ein besseres Verständnis der Vif-A3- und Vif-Cullin-Interaktionen birgt Potenzial für die Entwicklung neuer HIV-Therapeutika.
Diese Studie charakterisiert den durch FIV Vif-vermittelten Abbau von A3 und die restriktive Aktivität von felinem A3Z3 gegen FIV-Reporterviren. Es konnte festgestellt werden, dass das Vif-Protein des Puma-Lentivirus (PLV) Typ 1695 zwar feline A3Z2 und A3Z2Z3 abbauen kann, jedoch nicht A3Z3. Die Aminosäure 201 in Vif wurde als Schlüsselfaktor für die Interaktion mit A3Z3 identifiziert. Eine Mutation an dieser Position in PLV1695 Vif erklärt dessen Unfähigkeit A3Z3 zu antagonisieren. Das 201YRF203-Motiv in PLV Typ B (PLVB) Vif, welches auch in Vifs von FIV (Hauskatze), PLV Typ A (PLVA) und HIV-1 konserviert ist, erweist sich als entscheidend für die Interaktion für den Abbau von A3. Zusätzlich degradieren FIV und PLVA Vif A3Z3 über mittels Aminosäuren im N- und C-Terminus, während PLVB Vif ausschließlich auf seine C-terminale Bereiche angewiesen ist und die entsprechende N-terminalen Aminosäuren in der A3Z3-Degradation inaktiv bleiben.
Im Hinblick auf die Vif-Cullin-Interaktionen zeigt die Studie eine differenzielle Bindung feliner Vif-Proteine an CUL2 und CUL5. Ein konserviertes Interface, das die Bindung an CUL5 vermittelt, wurde sowohl in HIV-1- als auch in felinen Vifs identifiziert. Bemerkenswert ist, dass PLVB Vif sowohl CUL2- als auch CUL5-E3-Ligase-Komplexe rekrutiert, um A3Z3 abzubauen. Eine neuartige Interaktion zwischen HIV- und felinem Vif mit CUL1 wurde entdeckt, wobei die funktionelle Relevanz dieser Interaktion noch weiterer Untersuchungen bedarf.
Zusammenfassend bietet diese Arbeit eine neuartige Perspektive auf die Interaktionen zwischen lentiviralen Vif-Proteinen und den Wirtsfaktoren A3 sowie CUL. Die Ergebnisse eröffnen neue Ansätze zur Aufklärung der komplexen Dynamik von Interaktionen zwischen viralen und Wirtsproteinen.
