Beschreibung:COX-2 hat zahlreiche physiologische Aufgaben im menschlichen Organismus, ist jedoch auch an einigen pathophysiologischen Prozessen beteiligt: So gibt es Hinweise auf tumorpromovierende und karzinogene Wirkung der COX-2. Da UV-Strahlung Hauttumoren verursachen kann, sollte in der vorliegenden Arbeit ein Zusammenhang zwischen den UV-Qualitäten UVA- und UVB-Strahlung und der COX-2-Expression in HaCat-Zellen, immortalisierten menschlichen Keratinozyten, näher untersucht werden. Als ein mögliches Bindeglied zwischen UV-Bestrahlung und COX-2-Expression wird unter anderem ein Mitglied aus der Familie der MAP-Kinasen vermutet: Die p38-MAP-Kinase, so dass auch ihre Rolle bei der COX-2-Expression in der vorliegenden Arbeit näher untersucht werden sollte.
Die vorliegende Arbeit zeigt, dass UVB-Bestrahlung – im Gegensatz zu UVA-Bestrahlung – zu einer deutlichen Induktion der COX-2 führt, und zwar sowohl auf RNA-, als auch auf Proteinebene. Als Vermittler dieser Induktion wurde die p38-MAP-Kinase identifiziert: Eine Hemmung derselben vor Bestrahlung unterdrückte die vermehrte COX-2-Expression. Wesentlich zur vermehrten COX-2-Expression trägt bei, dass UVB-Bestrahlung zu einer langanhaltenden COX-2-mRNA-Stabilisierung führt – wohingegen eine Stabilisierung nach UVA-Bestrahlung nur sehr geringfügig nachzuweisen war. Die Stabilisierung ließ sich verhindern, indem die p38-MAPK vor Bestrahlung gehemmt wurde.
Insgesamt konnte in der vorliegenden Arbeit also gezeigt werden, dass – im Gegensatz zur UVA-Strahlung – vor allem UVB-Strahlung eine vermehrte COX-2-Expression bewirkt, und zwar durch Stabilisierung der COX-2-mRNA. Diese wird vermittelt durch eine Aktivierung der p38-MAP-Kinase.
