Beschreibungen:Der Oomycet Phytophthora infestans verursacht die Kraut- und Knollenfäule der Kartoffel und dadurch jährlich viele Millionen Euro Kosten, die sowohl Ernteausfälle als auch Kontrollmaßnahmen umfassen. Iprovalicarb und Dimethomorph sind zwei Oomyceten-spezifische Wirkstoffe die weithin zur Bekämpfung verschiedener Phytophthora-Spezies eingesetzt werden. Trotz intensiver Forschungen ist aber weder der genaue Wirkort noch der Wirkungsmechanismus beider Wirkstoffe bekannt. Aufgrund vorhergehender Untersuchungen wurde der Wirkort beider jedoch im Bereich der Zellwandsynthese vermutet. Eine Hypothese ist ein Effekt auf die Lokalisation der Zellwandbiosyntheseenzyme Cellulose-Synthase und Chitin-Synthase.
Zur in situ Immunolokalisation der beiden Enzyme wurden daher in dieser Arbeit spezifische Antikörper gegen verschiedene Domänen der Cellulose-Synthase und Chitin-Synthase hergestellt und die Lokalisation und Verteilung beider Enzyme vor und nach einer Behandlung mit Iprovalicarb und Dimethomorph mittels Immunofluoreszenzmikroskopie untersucht. Die Cellulose-Synthase konnte in der Plasmamembran von Hyphen in einer gleichmäßig punktförmig verteilten Anordnung um das gesamte Hyphenlumen lokalisiert werden. Eine Behandlung mit Iprovalicarb führte zu einer partiellen Agglomeration der Cellulose-Synthase, während Dimethomorph keinen Einfluss auf die Lokalisation des Enzyms hatte.
Die Chitin-Synthase wurde nur in der Keimungsspitze von Zoosporangien lokalisiert. Eine Behandlung mit Iprovalicarb und Dimethomorph führte zu keiner Änderung der Lokalisation dieses Enzyms. Diese Ergebnisse wurden durch eine Färbung von Chitin mit Texas Red-markiertem Wheat Germ Agglutinin bestätigt.
Mit Hilfe der spezifischen Antikörper wurde für beide Enzymkomplexe eine Lokalisation in Detergenz-resistenten Membranen (DRM) gezeigt. Da in anderen Eukaryoten DRM sterolreiche Lipid Rafts sind, wurde eine Färbung von Sterolen mit Filipin durchgeführt. Der Farbstoff lagerte sich gleichmäßig über den gesamten Bereich der Membran von Hyphen und Zoosporangien an. Eine Behandlung mit den Wirkstoffen Iprovalicarb und Dimethomorph führte jedoch zu keiner offensichtlichen Änderung in der Verteilung der Sterole.
Um weitere Einblicke in die Gemeinsamkeiten und Unterschiede des Wirkungsmechanismus von Iprovalicarb und Dimethomorph zu gewinnen, wurde eine quantitative und qualitative Analyse der Proteome von unbehandelten und in Gegenwart von Iprovalicarb, Dimethomorph bzw. Metalaxyl angezogenen Phytophthora infestans-Kulturen durchgeführt. Proteine, die im Vergleich zur unbehandelten Kontrolle ein verändertes Volumen aufwiesen, wurden sequenziert und durch einen Vergleich mit einer Phytophthora infestans Gendatenbank identifiziert. Während Iprovalicarb und Dimethomorph einige Ähnlichkeiten in den hervorgerufenen Effekten aufwiesen, unterscheidet sich der Effekt von Metalaxyl deutlich. Sowohl auf Iprovalicarb als auch auf Dimethomorph reagierte Phytophthora infestans mit einem generellen pflanzenähnlichen Abwehrmechanismus. Weitere Änderungen die durch Dimethomorph verursacht wurden, könnten als Fehler im Transport von Zellwandmaterial und dem daraus folgenden Versuch der Zellen überflüssige Glukane abzubauen, gedeutet werden.
Schließlich wurde die phylogenetische Verteilung der Cellulose-Synthase und Chitin-Synthase analysiert. Beide Enzyme sind sowohl in Eukaryoten als auch in Prokaryoten vertreten. Das Vorkommen von Cellulose-Synthase- und Chitin-Synthase-Genen in vielen Verwandtschaftsbereichen der Eukaryoten deutet darauf hin, dass beide Gene im letzten gemeinsamen Vorfahren der Eukaryoten vertreten waren und dieser wiederum möglicherweise eine Zellwand besaß.

The oomycete Phytophthora infestans not only causes potato late blight but, as a consequence, gives rise to costs of several million dollars annually including those incurred by crop losses and control measures. Iprovalicarb and dimethomorph are two oomycete-specific chemical components widely used against different Phytophthora species. Despite intensive research carried out over the last years, the target as well as the mode of action of both components remain unknown. Based on previous findings, the target was assumed to be involved in cell wall synthesis. One hypothesis is that these active ingredients affect the localisation of the cell wall synthesizing enzymes cellulose synthase and chitin synthase.
For in situ immunolocalisations of both enzymes, specific antibodies against different cytosolic domains of the cellulose synthase and the chitin synthase from Phytophthora infestans were produced. Then, by using immunofluorescence microscopy, the localization and distribution of both enzymes after treatment with iprovalicarb and dimethomorph were detected. Cellulose synthase was detected in the plasma membrane of Phytophthora infestans hyphae in a uniformly punctate distribution. Treatment with iprovalicarb led to a partial agglomeration of cellulose synthase, whereas dimethomorph had no effect on the localisation of the enzyme complex.
Chitin synthase was not detected in hyphae but in the tip of germinating zoosporangia. Treatment with iprovalicarb and dimethomorph did not impact the localisation of the chitin synthase. These results were improved by the staining of chitin with Texas Red-marked wheat germ agglutinin.
With the antibodies both enzymes were localised in detergent resistant membranes (DRM). In other eukaryotes, DRM contain sterol-rich lipid rafts and therefore Phytophthora infestans cells were stained with filipin. The dye bound uniformly to the whole membrane of hyphae and zoosporangia. Treatment with iprovalicarb and dimethomorph did not result in obvious changes in the distribution of sterols.
In order to gain insights into the similarities and differences of the modes of action of iprovalicarb and dimethomorph, quantitative and qualitative analyses of the proteome of untreated and chemical component-treated P. infestans cultures were performed. Proteins with altered levels in response to chemical component treatment compared to untreated controls were sequenced and their underlying genes were identified by comparison to the Phytophthora infestans genome. While iprovalicarb and dimethomorph reveal similarities in the produced effects, the effect of metalaxyl was clearly distinct from both other compounds. Phytophthora infestans exhibits a general plant-like defense response to iprovalicarb and dimethomorph. Furthermore, the changes caused by dimethomorph can possibly be ascribed to defects in the transport of cell wall materials and to a cellular attempt to degrade superfluous glucans.
Finally, the phylogenetic distribution of cellulose synthase and chitin synthase were analysed. Both enzymes were represented in eukaryotes and prokaryotes.
